In hostels, men and women may have to share a mixed dorm. Make sure your group is aware of this at the beginning of the tour. Likewise any mixed shower blocks.
Ask the group to stay on board the bus until you have sorted rooms out, as many reception areas are quite small. Arrange with the driver and the hotel beforehand what they want to do about unloading and storing bags while the reception process takes place.
Use the bus PA system to give out information rather than shout inside the hotel.
Some hotels offer a welcome drink to groups while they are waiting for rooms to be sorted out, it's worth asking the manager if he is willing to do this.
You may have to check - subtly- the relationship between older and younger single people of the opposite sex whose surname is different. They may be step-parent/stepchild, or they may be a couple.
The issue of double or twin beds may well need to be addressed - ask a couple of their preferences but never make any promises.
